In 1996 Capito won a Kanawha County seat in the West Virginia House of Delegates, where she stayed for two terms, until 2000.

That year she ran for the open 2nd congressional district seat vacated by Democrat Bob Wise and beat Democrat Jim Humphreys by a two-point margin, becoming the first Republican to represent the state in Congress since 1983; she won reelection through 2012 and held the seat from 2001 to 2015.

In 2014 she ran for the Senate seat of retiring Democrat Jay Rockefeller, won the Republican primary with 87% and beat Secretary of State Natalie Tennant 62% to 34%, becoming the first woman West Virginia sent to the U.S. Senate.

She won a second term in 2020 over Democrat Paula Jean Swearengin with more than 70% of the vote. Capito has led the Senate Committee on Environment and Public Works since 2025, and she is dean of West Virginia's congressional delegation.
