Before holding elected office, Mullin worked as district director to then-state senator Jackie Speier and as political director for his father. He also founded KM2 Communications, a multimedia production business in South San Francisco, and hosted programs on Peninsula-TV Channel 26. He was elected to the South San Francisco City Council in 2007 and served one term, including one year as mayor.

Mullin won a seat in the California State Assembly in 2012, representing the 22nd district, which covered most of the San Francisco Peninsula. He served as assistant speaker pro tempore during his first term and as speaker pro tempore from 2014 until he left office on December 5, 2022.

When Speier retired from the U.S. House, Mullin ran to succeed her in 2022 and defeated San Mateo County supervisor David Canepa with 55.8% of the vote. In the 119th Congress he sits on the Committee on Energy and Commerce.
